چکیده

Initiator methionine tRNA (tRNAIMet) species from wheat embryo has been purified by fractionation on a benzoylated DEAE-cellulose column followed by chromatography on a DEAE-Sephadex column. Transfer RNAIMet can be aminoacylated by the homologous enzyme as well as by Escherichia coli aminoacyl-tRNA synthetase. Unlike other eukaryotic cytoplasmic initiator tRNAs, the wheat embryo initiator tRNA cannot be formylated by E. coli transformylase. Amino acid incorporation studies in vitro using a wheat embryo ribosomal system and poly[r(A-U-G)] as messenger show that at low Mg2+ concentration polymethionine synthesis from Met-tRNAgMet (chain propagator species) is dependent on the presence of Met-tRNAIMet. Met-tRNAIMet cannot insert methionine internally in the homologous system but in a heterologous E. coli or rabbit reticulocyte ribosomal system Met-tRNAIMet appears to donate methionine into internal positions of the polypeptide chain. Analysis of oligonucleotides produced by digestion of tRNAIMet with ribonuclease T1 shows that the 5’and 3’-terminal nucleotide sequences are PA-U-C-A-Gand A-U-AC-A-AoH, respectively. As in other eukaryotic cytoplasmic initiator tRNAs the tetranucleotide sequence T-#-C-G(A)always present in loop IV of other tRNAs is absent. The tentative sequence of loop IV in the case of wheat embryo tRNAIMet is A-U*-C-G-mlA-A-A. Except for the presence of U* instead of U, this sequence is identical with those found in yeast, mouse myeloma, rabbit liver, and sheep mammary gland initiator tRNAs and different from that of E. coli initiator tRNA.

Amino acid incorporation and aminoacyl transfer in a wheat embryo system.

An amino acid-incorporating system has been obtained from ungerminated wheat embryos. This system has very low endogenous activity, but phenylalanine incorporation can be stimulated by the addition of polyuridylic acid. The poly U-stimulated system is similar to those from other sources in that it requires ribosomal particles, supernatant fluid, soluble ribonucleic acid, adenosine triphosphate,...

Methionine transfer ribonucleic acids of avian myeloblastosis virus.

Chick-embryo cells contain four isoaccepting species of methionine transfer RNA (I-IV). One species (I) is the initiator, tRNA(f) (Met), and the others (II, III, and IV) are the donors of internal methionyl residues (tRNA(m) (Met)). Over 85% of the tRNA(Met) in purified avian myeloblastosis virus consists of one tRNA(m) (Met) species, which resembles host-cell tRNA(Met) IV with respect to chrom...

Protein chain initiation by methionyl-tRNA in wheat embryo.

The mechanism of protein chain initiation has been investigated in a cell-free amino acid incorporation system from wheat embryos dependent on tobacco mosaic virus RNA. Analysis of the N-termini of the labeled peptide products of short-term incubations showed the presence of unblocked methionine. In addition, methionyl-tRNA (Met-tRNA) could be bound to ribosomes at 1.3 mM Mg(++) in a reaction r...

Characterization of two species of methionine transfer ribonucleic acid from bakers' yeast.

The biochemical properties of two species of methionine transfer ribonucleic acid from bakers’ yeast, separated by diethylaminoethyl Sephadex column chromatography, have been studied. Each species of the two methionine tRNAs, designated methionine tRNA I and methionine tRNA II, respectively, has been purified by benzoylated DEAEcellulose column chromatography. Methionine tRNA I can be esterifie...
